Application of isothermal titration calorimetry for detecting lipid membrane solubilization

چکیده
Isothermal titration calorimetry is presented to be an excellent method of detecting lipid membrane solubilization upon the addition of detergents. As an example, the titration of the nan-ionic detergent C t2EOt, to the lipid POPC is studied. The main contribution to the measured titration heat is the detergent's enthalpy difference upon transfer between micelles and membranes. The detergent added as micelles is transferred into membranes up to the saturating detergent per lipid mol/mol ratio, Rsa t = 1.45 + 0.07. Beyond Rsa t the membranes are solubilized to micelles. Subsequently, the titration heat changes its sign from endothermic to exothermic. After complete membrane dissolution at Rso ~ = 5.0 + 0.4 the titration heat vanishes nearly completely.

Triton promotes domain formation in lipid raft mixtures.
Biological membranes are supposed to contain functional domains (lipid rafts) made up in particular of sphingomyelin and cholesterol, glycolipids, and certain proteins. It is often assumed that the application of the detergent Triton at 4 degrees C allows the isolation of these rafts as a detergent-resistant membrane fraction. The current study aims to clarify whether and how Triton changes the...

Most of the biological phenomena are influenced by intermolecular recognition and interaction. Thus, understanding the thermodynamics of biomacromolecule ligand interaction is a very interesting area in biochemistry and biotechnology. One of the most powerful techniques to obtain precise information about the energetics of (bio) molecules binding to other biological macromolecules is isoth...

Understanding the structure, folding, and interaction of membrane proteins requires experimental tools to quantify the association of transmembrane (TM) helices. Here, we introduce isothermal titration calorimetry (ITC) to measure integrin αIIbβ3 TM complex affinity, to study the consequences of helix-helix preorientation in lipid bilayers, and to examine protein-induced lipid reorganization. P...

Interaction of curcumin with lipid bilayers is not well understood. A recent experiment showed that curcumin significantly affected the single-channel lifetime of gramicidin in a 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC) bilayer without affecting its single-channel conductance. We performed two experiments to understand this result. By isothermal titration calorimetry, we measured the par...

MICROCALORIMETRY Introduction Many industrial and biochemical applications depend on the usage of detergents and other surfactants. These compounds are indispensable, in particular, for the extraction, purification, and handling of membrane proteins. Owing to their amphiphilic nature, detergents can provide a membrane-mimetic environment required by integral membrane proteins to retain their na...
